A cell surface‐reducing microenvironment induces early osteogenic commitment

چکیده

Stem cell-based therapy has been highlighted as a potential avenue to promote tissue regeneration, where stimulation of stem cells differentiate into the targeted cell type is essential. One factors that induce their surrounding microenvironment. In this study, correlation between mild reductant and early osteogenic commitment was evaluated. A surface-reducing microenvironment significantly silenced transforming growth factor (TGF)-β signaling pathway mesenchymal (MSCs), followed by increased focal adhesion inhibition membrane protein dimerization. Furthermore, in vivo transplantation MSCs exposed reducing resulted an neobone formation. Thus, these results highlight influence commitment.

Microenvironment Modulates Osteogenic Cell Lineage Commitment in Differentiated Embryonic Stem Cells

BACKGROUND Due to their self-renewal, embryonic stem cells (ESCs) are attractive cells for applications in regenerative medicine and tissue engineering. Although ESC differentiation has been used as a platform for generating bone in vitro and in vivo, the results have been unsatisfactory at best. It is possible that the traditional culture methods, which have been used, are not optimal and that...

GATA-3 dose-dependent checkpoints in early T cell commitment.

GATA-3 expression is crucial for T cell development and peaks during commitment to the T cell lineage, midway through the CD4(-)CD8(-) (double-negative [DN]) stages 1-3. We used RNA interference and conditional deletion to reduce GATA-3 protein acutely at specific points during T cell differentiation in vitro. Even moderate GATA-3 reduction killed DN1 cells, delayed progression to the DN2 stage...

STAT3 as a Key Factor in Tumor Microenvironment and Cancer Stem Cell

Background Recent studies revealed that tumor-associated macrophages (TAMs) play a decisive role in the regulation of tumor progression by manipulating tumor oncogenesis, angiogenesis and immune functions within tumor microenvironments. Signal transducer and activator of transcription 3 (STAT3), which is a point of convergence for numerous oncogenic signalling pathways, is constitutively activ...

Single cell analysis of commitment in early embryogenesis.

Fate maps of amphibian embryos tell us the destination of certain areas at later stages of development. After studying Vogt's fate maps, Spemann wrote (in 1938) that "the question which at once calls for an answer is whether this pattern of presumptive primordia in the beginning gastrula is the expression of a real difference of these parts, whether they are already more or less predestined or ...

Umbelliprenin Induces Apoptosis in CLL Cell Lines

Chronic lymphocytic leukemia (CLL) remains an incurable disease that requires innovative new approaches to improve therapeutic outcome. Many Ferula species, including F. asa-foetida, synthesize terpenyloxy coumarins. One of these coumarins is umbelliprenin, which has been implicated with induction of apoptosis in some cancer cell lines.     In this study induction of apoptosis by umbellipre...
